Passive transfer of serum from animals vaccinated with RSV, Vac-F, Vac-G, and Bac-FG did NOT lead to potentiated disease following RSV challenge. RSV/form was prepared by zonal centrifugation, which concentrated the virus approximately 100X. [Note that SKB said Upjohn told them this Bac-FG was "old".] In the first of their 2 studies, Bac-FG and F, although inducing changes, did not reach statistical significance. Highest ELISA/neut rations wre with F and Bac-FG. However, in study 2, highest pathology was in Bac-FG (ELIAS/neut = 119) and 1:25 RSV/form (ONLY 17); while Vac-F, which showed NO pathology, had ratio of 15. No significant difference in pathology between RSV/form and PIV3/form. Passive transfer of antiserum to RSV, RSV/form, Bac-FG, Vac-F and Vac-G, to achieve circulating titers equivalent to post-vaccination, did not cause enhanced pathology--although no positive control was included. HOWEVER, whereas active immunization with Bac-FG reduced lung virus 103.8, passive immunization reduced it only 100.6--THUS, to rule out an Arthus reaction on the basis of the failure to duplicate the effect by passive transfer of serum is insufficient evidence. [However, reduction in pulmonary titer with RSV/form serum--102.4--was identical to that achieved with active immunization, but did not cause pathology.] Magnitude of pathology did not correlate, either directly or inversely, with magnitude of viral replication.
